目的探讨黄连素联合谷氨酰胺对癌性恶病质模型的改善作用及机制。方法使用小白鼠结肠腺癌细胞株CT-26皮下注射BALB/c小白鼠,构建荷瘤小白鼠肿瘤恶病质模型,将小白鼠分为5组:A组(无荷瘤小白鼠组)、B组(荷瘤无干预组)、C组(荷瘤黄连素干预组)、D组(荷瘤谷氨酰胺干预组)和E组(荷瘤黄连素联合谷氨酰胺干预组)。C组小白鼠给予黄连素灌胃[25 mg/(kg·d)],D组小白鼠给予谷氨酰胺灌胃[25 mg/(kg·d)],E组给予黄连素[25 mg/(kg·d)]联合谷氨酰胺[25 mg/(kg·d)]灌胃。连续2周记录小白鼠饮食量、体质量,测量并比较各组肿瘤瘤体质量、生化指标、血清白细胞介素1(interleukin 1,IL-1)、白细胞介素6(interleukin 6,IL-6)、肿瘤坏死因子(tumor necrosis factor,TNF)差异,观察黄连素联合谷氨酰胺对癌性恶病质疾病进展的干预效果。结果 C组、D组和E组相对于B组可显著改善荷瘤小白鼠的体质量减轻程度,改善生化指标,并降低血清中细胞因子IL-1、IL-6及TNF表达水平,差异有统计学意义(P<0.05)。其中E组指标更优于C、D组。结论口服黄连素联合谷氨酰胺可以有效改善癌性恶病质小鼠状态,其机制可能与降低细胞因子表达水平有关,需进一步研究。
Objective To investigate the effect and mechanism of berberine combined with glutamine on cancerous cachexia model. Methods BALB / c mice were injected subcutaneously with CT-26 mouse colon adenocarcinoma cell line to construct tumor cachexia model in mice. The mice were divided into 5 groups: group A (without tumor-bearing mice), group B (Tumor-free intervention group), group C (tumor-bearing berberine intervention group), group D (tumor-bearing glutamine intervention group) and group E (tumor bearing berberine combined with glutamine intervention group). The mice in group C were given gavage [25 mg / (kg · d)], and the mice in group D were given glutamine gavage [25 mg / (kg · d)]. The rats in group E were given berberine [25 mg / (kg · d)] combined glutamine [25 mg / (kg · d)] intragastric administration. The food intake and body weight of mice were recorded for 2 weeks. The body mass, biochemical indexes, serum levels of interleukin 1 (IL-1), interleukin 6 (IL-6) ) And tumor necrosis factor (TNF), and observed the intervention effect of berberine combined with glutamine on the progression of cancerous cachexia. Results Compared with group B, group C, group D and group E could significantly improve the degree of body weight loss, improve the biochemical indexes and decrease the expression levels of IL-1, IL-6 and TNF in the tumor-bearing mice Statistical significance (P <0.05). Among them, the index of group E is better than that of group C and D. Conclusions Oral administration of berberine combined with glutamine can effectively improve the status of cancerous cachexia mice. The mechanism may be related to the decrease of the expression of cytokines, which needs further study.
