目的探讨β-淀粉样蛋白25~35(amyloid-beta protein25~35,Aβ25~35)对体外培养血管平滑肌细胞(VSMC)的毒性作用。方法实验分对照组(生理盐水组)和Aβ25~35组(浓度分别为10、20、50、100μmol/L),分别培养1、3、10d,光镜下观察VSMC形态学改变;MTT法检测细胞活力;检测乳酸脱氢酶(LDH)活力。结果VSMC在浓度为20μmol/L的Aβ25~35孵育3d后即出现退行性改变,细胞存活率下降,LDH活力增高,其程度均呈Aβ25~35浓度、孵育时间依赖性。结论Aβ25~35可致VSMC形态学改变、存活率降低、LDH活力增高,有细胞毒性作用。
Objective To investigate the toxic effects of β-amyloid-beta protein 25-35 and Aβ25-35 on cultured vascular smooth muscle cells (VSMC). Methods The experiment was divided into control group (normal saline group) and Aβ 25-35 group (concentration of 10, 20, 50 and 100μmol / L respectively) for 1, 3 and 10 days. Morphological changes of VSMC were observed under light microscope. MTT assay Cell viability; detection of lactate dehydrogenase (LDH) activity. Results After VSMCs were treated with 20 μmol / L Aβ25-35 for 3 days, the VSMCs degenerated. The cell viability decreased and the LDH activity increased. The levels of Aβ25-35 were dependent on the incubation time. Conclusion Aβ25 ~ 35 can cause VSMC morphological changes, decreased survival, LDH activity increased, with cytotoxicity.
