目的探讨老年大鼠肝脏枯否细胞的前体细胞即外周血单核细胞对内毒素的反应性。方法将不同浓度脂多糖(内毒素)加入经培养的6及24月龄大鼠单核细胞中,2小时后检测单核细胞细胞骨架中肌动蛋白、肌球蛋白、波形蛋白等成分的含量。结果老年大鼠单核细胞骨架成分的含量显著低于年轻大鼠(P<0.01)。低浓度脂多糖使单核细胞骨架成分的含量增加,以年轻大鼠更为显著(P<0.01);高浓度脂多糖使单核细胞骨架成分的含量减少,以老年大鼠更显著(P<0.01)。结论老年大鼠单核细胞细胞骨架老化明显,更易受内毒素的损伤。
Objective To investigate the reactivity of the precursor cells of Kupffer cells in peripheral blood of aged rats with peripheral blood mononuclear cells (LPS) to endotoxin. Methods Different concentrations of lipopolysaccharide (endotoxin) were added into cultured monocytes of 6- and 24-month-old rats, and the contents of actin, myosin and vimentin in mononuclear cytoskeleton were detected 2 hours later . Results The content of monocyte cytoskeleton in aged rats was significantly lower than that in young rats (P <0.01). Low concentration lipopolysaccharide increased the content of monocyte cytoskeletal component, especially in young rats (P <0.01). The content of monocyte cytoskeleton decreased with high concentration of lipopolysaccharide, especially in the aged rats P <0.01). Conclusion The aging rat monocyte cytoskeleton aging significantly more vulnerable to endotoxin damage.
