目的探讨蛋白前体转化酶Furin和Fyn在宫颈癌组织中的表达及临床意义。方法常规方法提取41例宫颈癌和12例正常宫颈上皮的RNA及蛋白,RT-PCR方法检测Furin和Fyn mRNA表达水平,Western Blot检测二者蛋白表达水平。收集相关病理学资料进行统计学分析并研究二者蛋白表达与临床分期、病理分级和淋巴结转移等临床病理因素的关系。结果宫颈癌组织Furin和Fyn的mRNA表达及蛋白表达水平均明显高于正常宫颈组织,二者在宫颈癌组织中的蛋白阳性表达率分别为58.5%和65.9%,其表达均与临床分期、浸润深度及淋巴结转移有显著相关性(P<0.05)。Furin与Fyn的表达呈正相关(r=0.438,P<0.01)。结论 Furin和Fyn与宫颈癌的发生、发展有关,可作为宫颈癌诊断及临床监测的分子标志物。
Objective To investigate the expression and clinical significance of Furin and Fyn in patients with cervical cancer. Methods The RNA and protein of 41 cases of cervical cancer and 12 cases of normal cervical epithelium were extracted by routine method. The mRNA expression of Furin and Fyn was detected by RT-PCR and Western blotting respectively. Collect relevant pathological data for statistical analysis and study the relationship between the protein expression and clinical pathological factors such as clinical stage, pathological grade and lymph node metastasis. Results The mRNA and protein expressions of Furin and Fyn in cervical cancer tissues were significantly higher than those in normal cervical tissues. The positive rate of protein expression in cervical cancer was 58.5% and 65.9%, respectively. The expression of Furin and Fyn was correlated with clinical stage, Depth and lymph node metastasis (P <0.05). The expression of Furin and Fyn was positively correlated (r = 0.438, P <0.01). Conclusion Furin and Fyn are related to the occurrence and development of cervical cancer and can be used as molecular markers for diagnosis and clinical monitoring of cervical cancer.
