目的:比较抗菌肽LL-37及其衍生物对肝癌细胞HepG-2增殖和凋亡的影响。方法:以肝癌细胞HepG-2为靶细胞,采用不同浓度抗菌肽LL-37及其衍生物处理后,通过四甲基偶氮噻唑蓝(MTT)比色法检测细胞的增殖情况,流式细胞术Annexin V/PI双染色测定细胞的凋亡情况。结果:随着抗菌肽LL-37及其衍生物作用浓度的增加和作用时间的延长,其对肝癌细胞增殖的抑制作用逐渐增强,二者的浓度都达到300μg/m L时作用72 h,对肝癌细胞增殖的抑制作用最大(P<0.01),且抗菌肽LL-37衍生物对肝癌细胞增殖的抑制率高于抗菌肽LL-37。流式检测结果显示:抗菌肽LL-37衍生物诱导的细胞凋亡率高于抗菌肽LL-37。结论:抗菌肽LL-37衍生物在抗肝癌细胞增殖和促进其凋亡方面的作用优于抗菌肽LL-37。
Objective: To compare the effects of antimicrobial peptide LL-37 and its derivatives on proliferation and apoptosis of HepG2 cells. Methods: HepG2 cells were treated with various concentrations of antimicrobial peptide LL-37 and its derivatives. The proliferation of HepG2 cells was detected by MTT assay. Flow cytometry Cell apoptosis was detected by Annexin V / PI double staining. Results: The inhibitory effect of LL-37 and its derivatives on the proliferation of hepatocellular carcinoma cells was enhanced with the increase of the concentration and the action time of LL-37 and its derivatives. The concentration of LL-37 and its derivative The inhibition of proliferation of hepatocarcinoma cells was the largest (P <0.01), and the inhibitory rate of LL-37 derivative was higher than that of LL-37. The results of flow cytometry showed that the antiapoptotic LL-37 derivative induced higher apoptosis rate than the antimicrobial peptide LL-37. CONCLUSION: The antimicrobial peptide LL-37 derivative is superior to antimicrobial peptide LL-37 in inhibiting the proliferation and promoting apoptosis of hepatoma cells.
