本文叙述用1-羟基苯并三唑-氯化汞作柱前衍生的HPLC方法以测定霉菌发酵液中的天然青霉素。为了得到稳定的衍生物,含氨基青霉素有必要用苯甲酸酐先行酰化。缓冲液和衍生试剂配制 1-羟基苯并三唑6.76 g溶于10 ml水中,加入氯化汞溶液2.5 ml,然后用4M氢氧化钠溶液调节pH至9.2。最后体积为25 ml,1-羟基苯并三唑浓度为2M。苯甲酸酐0.45 g溶解于10 ml乙腈中得到0.2M溶液。
This paper describes the HPLC method using 1-hydroxybenzotriazole-mercuric chloride for pre-column derivatization to determine native penicillin in mold fermentation broth. In order to obtain a stable derivative, aminopenicillin must be acylated first with benzoic anhydride. Buffer and Derivatizing Agent 6.76 g of 1-hydroxybenzotriazole was dissolved in 10 ml of water, 2.5 ml of mercuric chloride solution was added, and the pH was adjusted to 9.2 with 4 M sodium hydroxide solution. The final volume was 25 ml and the 1-hydroxybenzotriazole concentration was 2M. Benzoic anhydride 0.45 g was dissolved in 10 ml of acetonitrile to give a 0.2 M solution.
