目的探讨神经生长因子(NGF)和血管内皮生长因子(VEGF)在上皮性卵巢癌中的表达及相关性。方法选择2012-2015年在杭州市第一人民医院行手术切除并经病理确诊为上皮性卵巢癌标本60例设为研究组,19例正常卵巢组织设为对照组,使用免疫组织化学法分别检测两组中NGF及VEGF的表达情况;构建NGF shRNA表达载体,使其能够抑制细胞内NGF基因表达,将经NGF shRNA基因下调后的细胞株与阴性对照细胞株(NC)、正常细胞株(normal)进行对照实验,通过Western-blot分别检测干扰前后卵巢癌细胞系A2780中NGF及VEGF蛋白质的表达情况。结果研究组NGF及VEGF蛋白阳性率均为100%,高于对照组中NGF及VEGF蛋白阳性率74%、84%,差异有统计学意义(P=0.004、0.027)。Western-blotting检测到NGF shRNA细胞表达株中NGF、VEGF蛋白水平的抑制率分别为40%、38%。NGF shRNA组中NGF与VEGF蛋白表达显著相关,差异有统计学意义。结论 NGF及VEGF对上皮性卵巢癌的发生、增殖及侵袭等有一定促进作用。
Objective To investigate the expression and correlation of nerve growth factor (NGF) and vascular endothelial growth factor (VEGF) in epithelial ovarian cancer. Methods Sixty patients with surgically diagnosed epithelial ovarian cancer who were surgically removed from the First People’s Hospital of Hangzhou City from 2012 to 2015 were selected as the study group and 19 normal ovarian tissues as the control group. Immunohistochemistry The expression of NGF and VEGF in both groups was established. The NGF shRNA expression vector was constructed to inhibit the expression of NGF gene in the two groups. The NGF shRNA knocked down the NCC and negative control cell lines (NC) and normal cell lines ) Were used as control. The protein expression of NGF and VEGF in ovarian cancer cell line A2780 before and after interference were detected by Western-blot. Results The positive rate of NGF and VEGF in study group was 100%, which was higher than that in control group (74%, 84%). The difference was statistically significant (P = 0.004, 0.027). Western-blotting detected NGF shRNA expression in cell lines NGF, VEGF protein levels were 40%, 38%. The expression of NGF and VEGF protein in NGF shRNA group was significantly correlated, the difference was statistically significant. Conclusion NGF and VEGF may promote the occurrence, proliferation and invasion of epithelial ovarian cancer.
