目的探讨甲苯暴露对小鼠肝脏、肺脏、肾脏、心脏、脑组织等靶点氧化损伤情况,探寻甲苯对机体作用的敏感、早期检测指标。方法将18只染毒小鼠根据体重随机分成甲苯高剂量组、低剂量组和对照组,采用静式吸入染毒方式,每天染毒2 h,共染毒30 d。实验结束后,测定小鼠各脏器中的氧化损伤指标。结果各脏器中SOD、GSH含量随染毒浓度的增加而减少(P<0.05),MDA含量随染毒浓度的增加而增加(P<0.05),染毒组和对照组的肺脏脏器系数明显不同(P<0.05),以上差异均有统计学意义。甲苯高剂量染毒组肝脏、心脏和肺脏中GSH、SOD含量均低于对照组(P<0.05),脑组织中GSH含量低于对照组(P<0.05),肾脏中GSH含量低于对照组(P<0.05);低剂量组心组织中SOD含量低于对照组(P<0.05);以上差异均具有统计学意义。结论不同浓度甲苯暴露下,小鼠各脏器均有氧化损伤;随着甲苯浓度的增加:小鼠肝脏、心脏、肺脏的GSH、SOD含量显著降低,MDA含量显著升高,且肝脏最为敏感;小鼠肾脏、脑组织的GSH含量逐渐降低,MDA含量逐渐升高。
OBJECTIVE To investigate the oxidative damage induced by toluene exposure in liver, lung, kidney, heart, brain and other targets of mice and explore the sensitive and early detection indexes of toluene on the body. Methods Eighteen infected mice were randomly divided into high dose of toluene group, low dose group and control group according to their body weight. The animals were exposed to inhalation for 2 hours daily for 30 days. After the experiment, the indicators of oxidative damage in various organs of mice were determined. Results The content of SOD and GSH in various organs decreased with the increase of the concentration of MDA (P <0.05), while the content of MDA increased with the increase of the concentration of MDA (P <0.05) Significantly different (P <0.05), the above differences were statistically significant. The levels of GSH and SOD in the liver, heart and lung of the high-dose toluene exposure group were lower than those in the control group (P <0.05), GSH content in the brain tissue was lower than that in the control group (P <0.05) (P <0.05). The content of SOD in the low-dose group was lower than that in the control group (P <0.05). All the above differences were statistically significant. CONCLUSION: Exposure of different concentrations of toluene to oxidative damage in all organs of mice. With the increase of toluene concentration, the contents of GSH and SOD in the liver, heart and lung of mice were significantly decreased, while the content of MDA was significantly increased and the liver was the most sensitive. Mouse kidney, brain GSH content gradually decreased, MDA content gradually increased.
