目的探讨氯乙烯(VCM)染毒对雄性大鼠肝组织及血清中mir-122表达量的影响。方法将无特定病原体级雄性SD大鼠分为对照组和低、中、高剂量组,每组24只。腹腔注射剂量分别为0、5、25和125 mg/kg体质量的VCM气体染毒,每周3次(隔日染毒)。在染毒第6、8和12周末,每组随机处死8只大鼠,分别提取肝组织中的小片段RNA(<200 nt)和血清中总RNA,采用实时荧光定量聚合酶链式反应检测mir-122相对表达水平。结果分别与同时间点对照组和低剂量组比较,中和高剂量组大鼠肝组织中mir-122相对表达水平在染毒第6周均上调(P<0.05),在染毒第12周均下调(P<0.05);中和高剂量组大鼠肝组织中mir-122相对表达水平均呈随时间增加而下降的趋势(P<0.05)。与同时间点对照组比较,3个剂量组大鼠血清中mir-122相对表达水平在染毒第8周均上调(P<0.05),在染毒第12周均下调(P<0.05);3个剂量组大鼠血清中mir-122相对表达水平在第12周均低于同组第6和8周(P<0.05)。结论 VCM可导致大鼠肝组织及血清中mir-122表达的改变;mir-122可能是VCM暴露所致的效应标志物之一。
Objective To investigate the effect of VCM on the expression of mir-122 in the liver and serum of male rats. Methods Male SD rats without specific pathogen were divided into control group and low, medium and high dose groups, 24 in each group. VCM gas doses of 0, 5, 25 and 125 mg / kg body weight were intraperitoneally administered 3 times a week (every other day). At the end of the 6th, the 8th and the 12th week of exposure, 8 rats were randomly sacrificed in each group. Small RNA (<200 nt) and total RNA in serum were extracted from the liver tissue and detected by real-time fluorescence quantitative polymerase chain reaction mir-122 relative expression level. Results Compared with the point-control group and the low-dose group at the same time, the relative expression level of mir-122 in the middle and high dose groups were all increased in the 6th week (P <0.05) (P <0.05). The relative expression levels of mir-122 in the middle and high dose groups of rats were decreased with the increase of time (P <0.05). Compared with the control group at the same time point, the relative expression levels of mir-122 in the three dose groups were all increased at the 8th week (P <0.05) and decreased at the 12th week (P <0.05). The relative expression level of mir-122 in serum of 3 dose groups was lower than that of the 6th and 8th week in the 12th week (P <0.05). Conclusions VCM can induce changes of mir-122 expression in rat liver tissue and serum; mir-122 may be one of the effect markers of VCM exposure.
