目的:雷公藤红素是一种天然的蛋白酶体抑制剂,有研究发现其对白血病细胞株、前列腺癌细胞株及多发性骨髓瘤细胞株均有抑制作用。我们研究其是否能抑制胃癌细胞株MGC803生长。方法:采用MTT增殖抑制试验、Hoechst 33258荧光染色、Western blot等方法,研究了雷公藤红素对胃癌细胞株OVCAR3的生长抑制作用。结果:雷公藤红素可以抑制胃癌细胞增殖,IC50值为4.69±0.38μM;通过Hoechst33258荧光染色、Caspase-3活化及PARP蛋白裂解,证实扁塑藤素通过诱导胃癌细胞凋亡作用,抑制其生长。结论:雷公藤红素通过诱导凋亡作用,抑制胃癌细胞生长,是一个有希望治疗胃癌的天然药物。
OBJECTIVE: Tripterine is a natural inhibitor of proteasome. Studies have shown that it inhibits leukemia cell lines, prostate cancer cell lines and multiple myeloma cell lines. We investigated whether it can inhibit the growth of gastric cancer cell line MGC803. Methods: MTT proliferation inhibition assay, Hoechst 33258 fluorescence staining, Western blot and other methods to study the tripterine on gastric cancer cell line OVCAR3 growth inhibition. Results: Tripterine inhibited the proliferation of gastric cancer cells with an IC50 value of 4.69 ± 0.38μM. Hoechst33258 fluorescence staining, Caspase-3 activation and PARP cleavage demonstrated that actinomycin inhibited the growth of gastric cancer cells by inducing apoptosis . Conclusion: Tripterine can inhibit the growth of gastric cancer cells by inducing apoptosis and is a natural drug promising to treat gastric cancer.
