目的:采用HPLC指纹图谱指导优化决明子有效组分纯化工艺。方法:采用HPLC建立决明子指纹图谱,以5个主要峰的回收率和指纹图谱相似度为指标,优选大孔树脂型号、上样量和洗脱条件。结果:D-101树脂对5个成分的回收率最高,上样量为3 m L,树脂用量为10 m L,洗脱溶剂为75%乙醇50 m L,纯化后主峰回收率均>85%,RSD均<0.6%,HPLC指纹图谱相似度>0.98。结论:采用HPLC指纹图谱指导优化决明子有效组分大孔树脂纯化工艺,可保持分离前后有效组分中化学成分的一致性。
Objective: To use HPLC fingerprinting to optimize the purification of effective components of cassia seed. Methods: The fingerprint of cassia seed was established by HPLC. The recovery of five main peaks and the similarity of fingerprint were taken as indexes. The selection of macroporous resin type, loading amount and elution conditions were optimized. Results: The highest recoveries of 5 components were obtained for D-101 resin with a loading volume of 3 mL, a resin volume of 10 mL, an eluting solvent of 75 mL of ethanol of 50 mL, and a peak recovery of> 85% , RSD <0.6%, HPLC fingerprinting similarity> 0.98. Conclusion: The HPLC fingerprint is used to guide the optimization of macroporous resin purification process of effective component of cassia seed, which can keep the consistency of the chemical components in the effective components before and after separation.
