本文采用脂多糖处理的RAW 264.7细胞为炎症细胞模型,采用ELISA、实时荧光定量PCR、免疫印迹、免疫细胞化学等方法探讨了覆盆子提取物对炎症发生过程中促炎、抗炎细胞因子及NF-κB的影响。结果表明,覆盆子提取物能够明显抑制脂多糖处理的RAW 264.7细胞中IL-1β、IL-6、NO、TNF-α和COX-2的表达(P<0.01或P<0.05),增强IL-10、HO-1等抗炎细胞因子的表达(p<0.01或P<0.05),并且提取物能够抑制NF-κB的产生。因此,我们推断覆盆子提取物具有较好的抗炎作用,其起效的可能机制是通过抑制NF-κB激活来抑制促炎因子的产生,并促进抗炎因子的表达。
In this study, RAW 264.7 cells treated with lipopolysaccharide (LPS) were used as inflammatory cell models. The effects of raspberry extract on proinflammatory, anti-inflammatory cytokines and NF in inflammatory process were investigated by using ELISA, real-time quantitative PCR, western blot and immunocytochemistry. -κB effect. The results showed that raspberry extract could significantly inhibit the expression of IL-1β, IL-6, NO, TNF-α and COX-2 in RAW 264.7 cells treated with lipopolysaccharide (P <0.01 or P < 10, HO-1 and other anti-inflammatory cytokines (p <0.01 or P <0.05), and the extract can inhibit the production of NF-κB. Therefore, we infer that raspberry extract has a good anti-inflammatory effect, and its possible mechanism is to inhibit the production of proinflammatory cytokines by inhibiting NF-κB activation and promote the expression of anti-inflammatory cytokines.
