目的 研究何首乌提取物对人乳腺癌MCF7细胞脂肪酸合酶 (fattyacidsynthase,FAS)的抑制作用。方法 MTT法测定人乳腺癌MCF7细胞增殖速度 ,采用超速离心技术部分纯化人乳腺癌MCF7细胞FAS。不同浓度何首乌提取物与FAS相互作用不同时间后 ,加入底物 ,用分光光度法观察何首乌提取物对FAS的抑制作用。结果 何首乌提取物对人乳腺癌MCF7细胞的增殖有一定的抑制作用。人乳腺癌MCF7细胞FAS被部分纯化 ,三种浓度何首乌提取物 (0 .1,0 .5 ,1g/L)与FAS在催化前相互作用 0、5和 10min ,对FAS活性的抑制率分别为 2 4 .6 0 %、2 7.30 %、and 4 2 .75 % (0 .1g/L) ;4 3.5 0 %、5 0 .30 %、and 94 .0 3% (0 .5 g/L) ;98.6 0 %、97.30 %and 97.0 5 % (1g/L)。结论 何首乌提取物对人乳腺癌MCF7细胞FAS具有抑制作用 ;作用强度既依赖于抑制剂浓度 ,又依赖于抑制剂与酶相互作用时间
Objective To study the inhibitory effect of Polygonum multiflorum Thunb extract on fatty acid synthase (FAS) in human breast cancer MCF7 cells. Methods MTT assay was used to determine the proliferation rate of human breast cancer MCF7 cells. FAS was partially purified from human breast cancer MCF7 cells by ultracentrifugation. Different concentrations of Polygonum multiflorum extract interact with FAS for different time, add substrate, and observe the inhibitory effect of Polygonum multiflorum extract on FAS by spectrophotometry. The results of Polygonum multiflorum extract on human breast cancer MCF7 cell proliferation to a certain extent. The FAS of human breast cancer MCF7 cells were partially purified. The three concentrations of Polygonum multiflorum extract (0 .1,0. 5, 1g / L) interact with FAS for 0, 5 and 10 min before catalysis, 4 3.5%, 50.0%, and 94.0% (0 .5 g / L) were found in the control group, ; 98.6 0%, 97.30% and 97.0 5% (1 g / L). Conclusion Polygonum multiflorum extract has inhibitory effect on FAS of human breast cancer MCF7 cells. The intensity of action depends not only on the concentration of inhibitor but also on the interaction time of inhibitor and enzyme
