亚硝基左旋精氨酸甲酯(L-NAME)是一氧化氮合成酶抑制剂,竞争性抑制体内一氧化氮(NO)合成。选择清洁级Wistar大鼠,随机分为3组,自妊娠第14天起分别皮下注射生理盐水、L-NAME 125mg/kg·d和L-NAME 250mg/kg·d,直到分娩,观察对孕鼠及胎鼠胎盘的影响。结果显示:皮下注射L-NAME可导致高血压、蛋白尿、肝肾功能及组织结构改变、胎鼠发育迟缓等妊娠高血压综合征的类似特征,并伴胎鼠后肢畸形,说明妊娠期NO合成减少可能是妊娠高血压综合征发病的一个重要因素。本方法为进一步研究建立了一个较理想的妊娠高血压综合征动物模型。
