本实验建立了一种浮动基点式 fura—2测定离体灌流心脏细胞内游离钙浓度的新模型。本模型中浮动基点式测定法克服了目前 fura—2技术中受细胞内某些物质产生的自身荧光变化干扰的缺点,实现了自动消除细胞自身荧光变化对实验结果的影响,并在此基础上将广泛用于测定游离状态的细胞内游离钙的 fura—2技术推广到了离体灌流心脏,实现了在更接近心肌生理状态的条件下测定其胞浆游离钙。
In this study, we established a new model of floating base-point fura-2 for determination of intracellular free calcium concentration in perfused heart. In this model, the floating basis point assay overcomes the shortcomings of the current fura-2 technology, which interferes with autofluorescence changes caused by certain substances in the cell and realizes the effect of automatically eliminating the change of cell autofluorescence on the experimental results. On the basis of this, The fura-2 technique, which is widely used to measure free intracellular free calcium, is extended to isolated perfused heart to measure the cytoplasmic free calcium in a condition that is closer to cardiac physiological state.
