目的 建立HPLC-MS/MS法测定老年房颤患者血浆中华法林对映异构体的浓度.方法 以ESI源负离子模式,萘普生为内标定量,华法林对映异构体和萘普生的检测离子对为(m/z)307.0→ 160.9和(m/z)和229.1→185.2,1mol· L-1盐酸作为缓冲环境,乙酸乙酯进行液液萃取.通过使用LuxCellulose-3手性色谱柱(2.1mm×100 mm,2μm)分离样品,流动相:水和甲醇(0.5%甲酸),流速:0.2 mL·min-1.结果 华法林对映异构体在0.1~10.0μg·mL-1内线性关系良好,定量下限为0.1 μg·mL-1.R-华法林批内和批间精密度的相对标准偏差(RSD)为4.99% ~8.13%和8.27%~ 13.41%,提取回收率85.90% ~97.70%.S-华法林批内和批间精密度的RSD为6.21%~699%和6.82%~14.99%,提取回收率82.40% ~ 93.90%.结论 本方法准确可靠,适用于老年房颤患者的华法林对映异构体血药浓度的分离检测.“,”Objective To establish a method for the quantitation of plasma warfarin enantiomer concentrations in elderly patients with atrial fibrillation using HPLC-MS/MS.Methods The mass spectrometer was operated in negative ESI model and used naproxen as internal standard,m/z 307.0→160.9 and 229.1→ 185.2 was used for the measurement of warfarin enantiomer and naproxen (IS),respectively.With 1 mol · L-1 hydrochloric acid as the buffer environment,liquid-liquidextraction with ethyl acetate separation was performed by Lux Cellulose-3 column (100 mm × 2.1 mm,5 μm) and an isocratic solvent system of 95∶5 (v/v) methanol:0.5% foric acid and water,with a flow rate of 0.2 mL· min-1 Results Calibration curve was linear over the range of 0.1-10.0 μg mL-1.The lower limit of quantification was 0.1 μg· mL-1.The intra-and inter-day precision and relative standard deviation (RSD) of R-warfarin in plasma were 4.99%-8.13% and 8.27%-13.41%.the extraction recoveries of R-warfarin were 85.90%-97.7%.The intra-and inter-day precision relative standard deviation (RSD) of S-Warfarin in plasma were 6.21%-6.99%% and 6.82%-14.99%.the extraction recoveries of S-warfarin were 82.40%-93.90%.Conclusion The HPLC-MS/MS developed in this study is accurate and reliable,thus it can be applied for the quantitation of warfarin enantiomers in plasma of elderly patients with atrial fibrillation.
