存放方式对船载冰箱保存红细胞活性与天然免疫功能的影响

目的探讨船载条件下血袋摆放方式对保存红细胞活性和天然免疫功能的影响。方法常规制备红细胞悬液4袋,每袋等分为4份,1份为对照组,留存陆地血库冰箱进行直立贴放,其余3份为实验组,于任务当天置于同1船载储血冰箱内,分别给予垂挂固定(E1)、直立贴放(E2)和平铺码放(E3)等不同方式储存,任务结束后检测每袋血液的红细胞渗透脆性、补体调控蛋白表达以及天然免疫黏附功能。结果与对照组相比,平铺码放方式保存的红细胞CD35以及免疫黏附功能均显著降低,渗透脆性明显增高;垂挂固定和直立贴放储存的红细胞,上述指标没有显著改变。3种不同存放方式对红细胞表面补体调控蛋白CD55和CD59的表达没有明显影响。结论从天然免疫黏附功能的角度分析,船载冰箱保存血液不适宜采用平铺码放方式。 Objective To investigate the effect of blood bag placement on the preservation of erythrocyte activity and innate immune function under shipboard conditions. Methods Four bags of erythrocyte suspensions were routinely prepared. Each bag was divided into four equal parts. One part was the control group. The refrigerator was placed upright on the terrestrial blood bank. The other three were experimental groups. On the same day, (E1), upright placement (E2) and tile placement (E3), respectively. After the task was completed, the erythrocyte osmotic fragility, complement regulatory protein expression and innate immune adhesion function in each bag of blood were measured. Results Compared with the control group, CD35 and immunoadhesion of erythrocytes preserved by tiling were significantly decreased, and osmotic fragility was significantly increased. There was no significant change in the above indexes when the erythrocytes were suspended and erected. Three different storage methods had no significant effect on the expression of complement regulatory proteins CD55 and CD59 on erythrocytes. Conclusion From the point of view of the function of innate immunity adhesion, it is unsuitable to store the blood in the ship-borne refrigerator by using the method of laying tile.
