目的:探讨肝型脂肪酸结合蛋白(L-FABP)和脂肪酸合成酶(FAS)与乳腺浸润性导管癌发生发展的关系。方法:用半定量逆转录聚合酶链反应、免疫组织化学和West-ern blot等方法检测L-FABP、FAS和血管内皮生长因子(VEGF)在86例乳腺浸润性导管癌中的表达变化。结果:L-FABP和FAS主要分布于浸润性导管癌的腺泡和导管上皮细胞,L-FABP的mRNA和蛋白质的表达量在Ⅲ级浸润性导管癌的较Ⅰ、Ⅱ级明显上调(P<0.05)。相反,FAS的表达量在Ⅲ级浸润性导管癌的较Ⅰ、Ⅱ级明显下调,两者的表达无明显相关性(P>0.05)。但是,L-FABP的表达与VEGF呈明显相关性。结论:在Ⅲ级浸润性导管癌,随着FAS表达下调,L-FABP和VEGF的表达上调。
Objective: To investigate the relationship between the expression of hepatic fatty acid binding protein (L-FABP) and fatty acid synthase (FAS) and invasive ductal carcinomas in breast. Methods: The expressions of L-FABP, FAS and VEGF in 86 cases of invasive ductal carcinoma of the breast were detected by semi-quantitative reverse transcriptase-polymerase chain reaction, immunohistochemistry and western blot. Results: L-FABP and FAS mainly distributed in the acinar and ductal epithelial cells of invasive ductal carcinoma. The mRNA and protein expression of L-FABP were significantly up-regulated in grade Ⅲ invasive ductal carcinomas compared with grade Ⅰ and Ⅱ (P < 0.05). In contrast, the expression of FAS was significantly down-regulated in grade III and grade II invasive ductal carcinomas, with no significant correlation (P> 0.05). However, L-FABP expression was significantly correlated with VEGF. Conclusion: In grade III invasive ductal carcinoma, the expressions of L-FABP and VEGF are up-regulated with the down-regulation of FAS expression.
