卵磷酯—胆固醇—酰基转移酶(LCAT)是酯化高密度脂蛋白(HDL)胆固醇的关键酶,在血浆脂蛋白代谢中起着重要作用。最近几个实验室已将此酶从人和一些哺乳动物的血浆中纯化成单体。鸭血清脂蛋白的HDL含量很高,是研究HDL中胆固醇酯化的较好的模型。为了研究血清脂蛋白的代谢机制,我们从鸭血清中提取纯化了LCAT。方法、结果如下:取新鲜鸭血清,先对Ⅰ=0.1的磷酸盐缓冲液透析,再用硫酸葡聚糖和CaCl_2(终浓度分别为1.4%和0.1M)沉
Lecithin-cholesterol-acyltransferase (LCAT) is a key enzyme in the esterification of high-density lipoprotein (HDL) cholesterol and plays an important role in the metabolism of plasma lipoproteins. Several laboratories have recently purified this enzyme from human and some mammalian plasma into monomers. HDL levels of duck serum lipoproteins are high, a good model for studying cholesterol esterification in HDL. In order to study the metabolic mechanism of serum lipoproteins, we extracted and purified LCAT from duck serum. The results were as follows: Fresh duck serum was dialyzed against phosphate buffer solution Ⅰ = 0.1 and then precipitated with dextran sulfate and CaCl 2 (final concentrations of 1.4% and 0.1 M, respectively)
