采用硅胶柱色谱、Sephadex LH-20凝胶柱色谱和高效液相色谱等方法对白茅内生菌Chaetomium globosum固体发酵产物的化学成分进行分离纯化,获得9个化合物。利用高分辨质谱、核磁共振谱及文献比对的方法,将上述化合物分别鉴定为chaetoglobosin F(1),chaetoglobosin Fex(2),chaetoglobosin E(3),cytoglobosin A(4),penochalasin C(5),isochaetoglobosin D(6),N-benzoylphenylalaninyl-N-benzoylphenylalaninate(7),尿嘧啶(8),5-甲基尿嘧啶(9),其中化合物7为首次从毛壳菌中分离得到。采用MTT法测定化合物的体外细胞毒活性,结果显示,化合物1,3,4,5对人宫颈癌细胞株He La具有明显的抑制活性,IC50分别为99.43,23.77,97.92,86.25μmol·L~(-1),而阳性对照药顺铂的IC50为24.33μmol·L~(-1)。
The chemical constituents of the solid fermentation product of Chaetomium globosum were isolated and purified by silica gel column chromatography, Sephadex LH-20 gel column chromatography and high performance liquid chromatography. Nine compounds were obtained. The above compounds were identified as chaetoglobosin F (1), chaetoglobosin Fex (2), chaetoglobosin E (3), cytoglobosin A (4) and penochalasin C (5) respectively by high resolution mass spectrometry, nuclear magnetic resonance spectroscopy and literature comparison , isochaetoglobosin D (6), N-benzoylphenylalaninyl-N-benzoylphenylalaninate (7), uracil (8) and 5-methyluracil (9) were isolated from Chaetomium for the first time. The cytotoxic activities of the compounds were determined by MTT assay. The results showed that the compounds 1, 3, 4, and 5 have significant inhibitory activity on human cervical cancer cell line He La with IC50 of 99.43, 23.77, 97.92 and 86.25 μmol·L- (-1), while the positive control drug cisplatin IC50 was 24.33μmol·L -1.
