构建了含人血清白蛋白(hALB)基因的cDNA全序列、内含子1以及山羊β-酪蛋白基因启动子和5’端上游调控序列在内的乳腺表达载体pcDNA3.1-GCALBm,并通过显微注射将其导入经过体外受精(IVF)的奶牛受精卵中,体外培养到桑椹胚后期,取出少许细胞,应用巢式PCR对其进行目的基因整合及性别的预鉴定,将10枚整合胚移植于8头受体母牛,结果有3头妊娠,受孕率为37.5%(3/8),其中2头于妊娠中期流产,另有1头足月分娩产下1头有人血清白蛋白基因整合的雄性转基因试管牛,转基因实验的总有效率为12.5%(1/8)。
