目的:纹状体神经元可以投射突触到室管膜下区(SVZ),并影响SVZ区域前体细胞的活性,但是纹状体神经元活性如何影响SVZ区域神经再生未知,本文旨在探讨纹状体神经元与SVZ区域神经再生的关系。方法:利用光遗传学的方法,特异调控纹状体神经元活性后,利用免疫组织化学方法观察其活性对SVZ区域神经再生的影响,并且探讨其可能机制。结果:当以CaM KⅡ作为启动子时,光敏感蛋白可以在纹状体神经元中表达,而且纹状体神经元的发放频率能够被激光控制。纹状体神经元活性的抑制促进NT-1的分泌(P<0.05),免疫组织化学结果显示纹状体神经元活性的降低促进SVZ区域BrdU与DCX双阳性细胞数目的增多(P<0.05)。结论:纹状体神经元活性是影响SVZ区域神经再生的重要因素,抑制纹状体神经元活性可以促进神经再生。
OBJECTIVE: Striatum neurons project synaptically to the subventricular zone (SVZ) and affect the activity of SVZ precursors, but how neuronal activity in the striatum affects SVZ is unknown. This article aims to investigate Relationship between striatal neurons and nerve regeneration in SVZ area. Methods: Using the method of optogenetics to specifically regulate the activity of neurons in the striatum, the effect of its activity on the neurogenesis in the SVZ was observed by immunohistochemistry and the possible mechanism was explored. Results: When CaM KⅡ was used as a promoter, light-sensitive proteins could be expressed in striatum neurons and the firing frequency of striatal neurons could be controlled by laser. The inhibition of striatal neuronal activity promoted the secretion of NT-1 (P <0.05). Immunohistochemistry showed that the decrease of striatal neuronal activity promoted the increase of BrdU and DCX double positive cells in SVZ (P <0.05) . Conclusion: The activity of striatal neurons is an important factor affecting the regeneration of nerve in SVZ region. Inhibition of the activity of striatal neurons can promote the regeneration of nerve.
