目的探讨磷酸化的真核细胞翻译起始因子4E(p-eIF4E)和磷酸化的eIF4E结合蛋白1(p-4EBP1)与鼻咽癌(NPC)临床病理特征和预后的相关性。方法免疫组织化学染色法检测314例NPC组织中p-eIF4E和p-4EBP1蛋白表达,利用SPSS16.0软件分析p-eIF4E和p-4EBP1蛋白表达与NPC临床病理特征及预后的相关性。结果 NPC组织中存在p-eIF4E和p-4EBP1蛋白异常高表达,阳性表达率分别为74.2%(233/314)、72.9%(229/314);p-eIF4E和p-4EBP1蛋白表达在淋巴结转移NPC组织中阳性表达率显著高于无淋巴结转移NPC组织,但与性别、年龄、临床分期和组织类型无关;NPC组织中p-eIF4E与p-4EBP1蛋白表达呈正相关,且p-eIF4E和p-4EBP1蛋白阳性的NPC患者生存时间明显短于p-eIF4E和p-4EBP1阴性患者,是NPC判断预后的独立因素。结论 p-eIF4E和p-4EBP1蛋白过表达可能与NPC的发生、发展及不良预后密切相关,p-eIF4E和p-4EBP1有望成为NPC临床诊治和判断预后的分子标志物。
Objective To investigate the relationship between the phosphorylation of eIF4E and phospho-eIF4E-1 (p-4EBP1) and the clinicopathological features and prognosis of nasopharyngeal carcinoma (NPC). Methods The expressions of p-eIF4E and p-4EBP1 in 314 NPC tissues were detected by immunohistochemical staining. The correlation between the expression of p-eIF4E and p-4EBP1 protein and clinicopathological features and prognosis was analyzed by SPSS16.0 software. Results The abnormal expressions of p-eIF4E and p-4EBP1 were found in NPC tissues. The positive rates of p-eIF4E and p-4EBP1 were 74.2% (233/314) and 72.9% (229/314), respectively. The positive expression rate of p-eIF4E and p-4EBP1 in NPC tissues was significantly higher than that in NPC tissues without lymph node metastasis, but not in gender, age, clinical stage and tissue type. The expression of p-eIF4E and p- Patients with 4EBP1-positive NPC had significantly shorter survival times than patients with p-eIF4E and p-4EBP1-negative tumors and were independent prognostic factors for NPC. Conclusion The overexpression of p-eIF4E and p-4EBP1 may be closely related to the occurrence and development of NPC and the prognosis of NPC. P-eIF4E and p-4EBP1 are expected to be the molecular markers for the clinical diagnosis and prognosis of NPC.
