目的:探讨犬腹腔镜手术对肝细胞凋亡的影响。方法:实验犬16只随机分为2组,A组行腹腔镜子宫切除术,B组行常规开腹子宫切除术。观察术前、术中、手术结束后2h肝功能指标血清谷氨酸氨基转移酶(ALT)、天门冬氨酸氨基转移酶(AST)变化;观察术前、术后2h肝脏凋亡指数变化,实时荧光定量RT-PCR检测FasmRNA、N-mycmRNA、nm23H1mRNA、MTA1 mRNA表达。结果:开腹手术组术前、术中、术后肝功能比较差异无统计学意义(P>0.05),腹腔镜组手术后2h ALT、AST高于其他时间点及开腹手术组。两组术前术后凋亡指数、FasmRNA、N-myc mRNA、nm23H1 mRNA、MTA1 mRNA表达水平差异无统计学意义(P>0.05)。结论;腹腔镜犬子宫切除术存在一过性肝功能改变,对肝细胞凋亡及细胞转移基因表达无影响。
Objective: To investigate the effect of canine laparoscopic surgery on hepatocyte apoptosis. Methods: Twenty-six dogs were randomly divided into two groups. Group A received laparoscopic hysterectomy and group B received routine laparotomy. The changes of hepatic function indicators such as ALT and AST were observed before, during and after surgery. The changes of liver apoptosis indexes were observed before and 2 hours after operation. Real-time fluorescent quantitative RT-PCR detection FasmRNA, N-mycmRNA, nm23H1mRNA, MTA1 mRNA expression. Results: There was no significant difference in hepatic function before, during and after operation between two groups (P> 0.05). The ALT and AST of the laparoscopic group were significantly higher than those of the other two groups at lapse of 2 hours. There was no significant difference in apoptotic index, expression of FasmRNA, N-myc mRNA, nm23H1 mRNA and MTA1 mRNA between the two groups before operation (P> 0.05). Conclusions There is a transient change of hepatic function in laparoscopic canine hysterectomy, which has no effect on hepatocyte apoptosis and gene expression of cell metastasis.
