[目的]探讨白首乌C-21甾体总苷(CGB)体外抑制大鼠胶质瘤细胞(C6细胞)生长的作用,并初步探讨其作用机制。[方法]采用MTT法检测CGB对C6细胞活性的影响,倒置普通光学及荧光显微镜观察CGB对细胞核形态的影响,流式细胞术检测细胞凋亡率。[结果]CGB对C6细胞的生长呈浓度依赖性抑制;CGB 90mg/L处理48h后,C6细胞的凋亡率显著增高(P<0.01),细胞核出现固缩、断裂;CGB作用48h对体外培养8d的神经元毒性小。[结论]CGB能明显抑制体外培养的C6细胞的生长;诱导肿瘤细胞凋亡是其作用机制之一。
[Objective] To investigate the inhibitory effect of C-21 steroidal glycoside (CGB) on the growth of rat glioma cells (C6 cells) in vitro and its possible mechanism. [Method] The effect of CGB on the activity of C6 cells was detected by MTT assay. The effect of CGB on the morphology of nucleus was observed by inverted optical microscope and fluorescence microscope. The apoptosis rate was detected by flow cytometry. [Results] CGB inhibited the growth of C6 cells in a concentration-dependent manner. CGB at 90 mg / L for 48 h significantly increased the apoptosis rate of C6 cells (P <0.01) Neurons of 8d are less toxic. [Conclusion] CGB can significantly inhibit the growth of C6 cells cultured in vitro; induce apoptosis of tumor cells is one of its mechanisms.
