目的观察滋阴活血方剂对糖尿病大鼠牙周组织的病理改变及VEGF表达的影响,探讨滋阴活血方剂对糖尿病牙周组织的作用。方法选用40只雄性Wistar大鼠,随机分为3组:正常组(10只)、糖尿病组(15只),滋阴活血方剂组(中药组)(15只)。糖尿病组、中药组腹腔注射STZ溶液,制备糖尿病大鼠模型。自建立糖尿病模型第一天起,中药组每只大鼠3ml/天(含生药1g/ml)中药灌喂;所有大鼠正常饮食水。持续喂养12周。对牙周组织切片进行HE染色和VEGF免疫组化染色,观察牙周组织的病理变化和VEGF表达的改变。结果糖尿病模型建立后,糖尿病组空腹血糖值[21.51±2.44mmol/L]和中药组空腹血糖值[21.76±1.82mmol/L]均高于正常组[5.51±0.75 mmol/L](P<0.05)。糖尿病组、中药组空腹血糖均大于15mmol/L,糖尿病模型建立成功。滋阴活血方剂干预12周后,中药组血糖[13.36±3.30mmol/L]下降(P<0.05),并低于糖尿病组[21.3±4.85mmol/L](P<0.05),均高于正常组[5.49±0.49 mmol/L](P<0.05)。中药组血糖水平[(13.36±3.30)mmol/L]显著低于糖尿病组[(21.3±4.85)mmol/L](P<0.05)。VEGF免疫组化平均光密度值(OD)糖尿病组[113.33±18.12]低于正常组[143.68±14.15]和中药组[132.39±10.66](P<0.05);正常组与中药组相比差异无统计学意义无差异(P>0.05)。结论滋阴活血方剂可降低大鼠实验性高血糖水平,并使其牙周组织中的VEGF表达水平趋于正常。
Objective To observe the effect of Ziyinhuoxue prescription on pathological changes and expression of VEGF in periodontal tissues of diabetic rats and explore the effect of Ziyin Huoxue prescription on periodontal tissues of diabetic rats. Methods Forty male Wistar rats were randomly divided into three groups: normal group (n = 10), diabetic group (n = 15) and Ziyinhuoxue prescription group (traditional Chinese medicine group) (n = 15). Diabetic group and traditional Chinese medicine group were intraperitoneally injected with STZ solution to prepare diabetic rat model. From the first day of establishment of diabetic model, each rat in traditional Chinese medicine group was infused with Chinese medicine at a dose of 1 ml / day (containing crude drug 1 g / ml); all rats were given normal diet water. Continue feeding for 12 weeks. Periodontal tissue sections were stained with HE and immunohistochemical staining for VEGF to observe the pathological changes of periodontal tissues and the changes of VEGF expression. Results After the model of diabetes was established, fasting blood glucose (21.51 ± 2.44mmol / L) and fasting blood glucose (21.76 ± 1.82mmol / L) in diabetic group were significantly higher than those in normal group [5.51 ± 0.75 mmol / L] ). The fasting blood glucose of diabetes mellitus group and traditional Chinese medicine group were more than 15mmol / L, and the diabetic model was established successfully. After 12 weeks of intervention of Ziyinhuoxue prescription, the blood sugar of Chinese medicine group (13.36 ± 3.30mmol / L) decreased (P <0.05) and was lower than that of diabetic group [21.3 ± 4.85mmol / L] (P <0.05) Group [5.49 ± 0.49 mmol / L] (P <0.05).
