目的探讨温肾养血方对无排卵大鼠抑制素—激活素—卵泡抑素(inhibin-activin-follistatin,INH-ACT-FS)系统和性腺激素水平的影响。方法 76只9日龄大鼠于颈背部皮下一次性注射丙酸睾丸酮1.25mg/只(0.05mL/只)制备无排卵大鼠模型。将58只造模成功大鼠按随机数字表法分为模型组(10只)、西药组(12只)和温肾养血方大、中、小剂量(简称中药大、中、小剂量组,每组12只)。另选10只22日龄大鼠作为正常组。正常组和模型组每天给予蒸馏水灌胃。西药组给予枸橼酸氯米芬(0.58mg/100g)灌胃,连续5日。中药大、中、小剂量组分别给予温肾养血方5.2、2.6、1.3mg/100g灌胃,连续21日。采用放射免疫法检测大鼠促卵泡刺激素(follicle stimulating hormone,FSH)、促黄体生成素(luteinizing hormone,LH)、雌二醇(estradiol,E2)、孕酮(progesterone,P)和催乳素(prolactin,PRL)水平;酶免法检测抑制素(inhibin,INH)、激活素(activin,ACT)和卵泡抑素(follistatin,FS)含量。结果与正常组比较,模型组大鼠血清FSH、LH升高,P降低,差异均有统计学意义(P<0.05);模型组INH降低,FS升高,差异有统计学意义(P<0.05)。与模型组比较,西药组和中药大、中、小剂量组大鼠血清FSH均下降,西药组P下降,差异均有统计学意义(P<0.05);西药组和中药大、中、小剂量组INH均升高,FS均降低,中药大剂量组ACT升高,差异均有统计学意义(P<0.05)。结论温肾养血方可能是通过调节INH-ACT-FS系统和性腺激素水平促进卵泡发育的。
Objective To investigate the effects of Wenshen Yangxue Prescription on inhibin-activin-follistatin (INH-ACT-FS) and gonadal hormones in anovulatory rats. Methods A total of 76 rats of 9 days old were injected subcutaneously with 1.25 mg / kg of testosterone propionate (0.05 mL / mouse) to establish an anovulatory rat model. Fifty-eight rats were randomly divided into model group (n = 10), western medicine group (n = 12) and Wenshen Yangxue Fang large, medium and small dose , 12 in each group). Another 10 22-day-old rats as a normal group. Normal group and model group were given distilled water daily. Western medicine group were given clomiphene citrate (0.58mg / 100g) gavage for 5 consecutive days. Traditional Chinese medicine large, medium and small dose groups were given Wenshen Yangxue Fang 5.2,2.6,1.3 mg / 100g gavage for 21 consecutive days. The levels of follicle stimulating hormone (FSH), luteinizing hormone (LH), estradiol (E2), progesterone (P) and prolactin prolactin and PRL levels were detected by enzyme-linked immunosorbent assay (ELISA). The levels of inhibin (INH), activin (ACT) and follistatin (FS) Results Compared with the normal group, the serum FSH, LH and P in the model group were significantly lower than those in the normal group (P <0.05); INH decreased and FS increased in the model group (P <0.05) ). Compared with the model group, serum FSH of Western medicine group and Chinese medicine large, medium and small dose groups decreased, and P of Western medicine group decreased (P <0.05); Western medicine group and traditional Chinese medicine large, medium and small dose Group INH were increased, FS decreased, high-dose Chinese medicine group ACT increased, the difference was statistically significant (P <0.05). Conclusion Wenshen Yangxue Prescription can promote follicular development by regulating INH-ACT-FS system and gonadal hormone levels.
