目的 :探讨转化生长因子 β(TGF - β)蛋白及mRNA基因在膜性肾小球肾炎中的表达及其在发病中的作用。方法 :采用常规病理、免疫组织化学及原位杂交方法对人膜性肾小球肾炎组织进行染色 ,并经医学图像分析系统进行分析。结果 :在正常对照组 ,TGF - β蛋白及TGF - β1mRNA于肾小管上皮细胞呈极微弱表达 ,肾小球内TGF- β蛋白及mRNA未见阳性表达。在膜性肾小球肾炎组织中 ,TGF - β蛋白在肾小管上皮细胞胞浆内呈强阳性表达 ,肾小球呈阳性表达 ,肾小球与肾小管阳性表达与正常对照组相比 (P <0 .0 1) ,均具有显著性差异。TGF - β1mRNA阳性表达主要位于肾小管上皮细胞胞浆内 ,与正常对照组相比 ,有显著性差异 (P >0 .0 5 ) ;肾小球内弱阳性表达 ,与正常对照组相比 ,无显著性差异 (P >0 .0 5 )。结论 :膜性肾小球肾炎时TGF - β蛋白在肾小球与肾小管表达均显著增高 ,TGF - β1mRNA在肾小管表达增高 ,提高TGF - β在肾小球硬化及肾小管间质纤维化中起重要作用。
Objective: To investigate the expression of transforming growth factor β (TGF - β) protein and mRNA in membranous glomerulonephritis and its role in the pathogenesis. Methods: Human membranous glomerulonephritis was stained by routine pathology, immunohistochemistry and in situ hybridization and analyzed by medical image analysis system. Results: In normal control group, TGF - β protein and TGF - β1 mRNA were very weakly expressed in renal tubular epithelial cells, and no expression of TGF - β protein and mRNA in glomeruli was found. In membranous glomerulonephritis tissues, TGF - βprotein was strongly expressed in the cytoplasm of renal tubular epithelial cells, glomerulus was positive, and glomerular and tubulocytosis was positively expressed compared with the normal control group (P <0. 01), all with significant differences. The positive expression of TGF - β1 mRNA mainly located in the cytoplasm of renal tubular epithelial cells, compared with the normal control group, there was a significant difference (P> 0.05); glomerular weak positive expression compared with the normal control group, No significant difference (P> 0.05). Conclusion: The expression of TGF - βprotein in glomerulus and tubule is significantly increased, the expression of TGF - β1mRNA is increased in tubulointerstitial of membranous glomerulonephritis and the expression of TGF - β is increased in glomerulosclerosis and tubulointerstitial fibrosis Play an important role.
