目的:观察天麻钩藤饮对血管紧张素Ⅱ(AngⅡ)致人脐静脉内皮细胞(HUVECs)损伤的保护作用。方法:制备大鼠天麻钩藤饮含药血清。采用酶消化法体外培养HUVECs,利用Human von Willebrand factor免疫细胞化学法鉴定,随机分为3组:对照组,AngⅡ组,AngⅡ+天麻钩藤饮组。倒置显微镜下观察细胞形态、密度,酶联免疫法测定细胞上清TNF-α含量,RT-PCR法测定细胞PPAR-γmRNA的表达。结果:与对照组比较,AngⅡ(10-6mol/L)可引起内皮细胞密度降低,细胞分泌TNF-α增加,PPAR-γmRNA表达水平降低。天麻钩藤饮含药血清可抑制AngⅡ导致的细胞损伤,减少TNF-α的分泌,升高PPAR-γmRNA的表达。结论:天麻钩藤饮可对抗AngⅡ所致的HUVECs损伤,保护血管内皮细胞功能。
Objective: To observe the protective effect of Tianma Gouteng Decoction on damage of human umbilical vein endothelial cells (HUVECs) induced by angiotensin II (AngII). METHODS: Rat serum containing Tianma Gouteng Decoction was prepared. HUVECs were cultured in vitro by enzyme digestion and identified by immunocytochemistry using Human von Willebrand factor. They were randomly divided into three groups: control group, AngII group, and AngII+ Tianma Gouteng drink group. The morphology and density of the cells were observed under an inverted microscope. The content of TNF-α in the supernatant was determined by enzyme-linked immunosorbent assay. The expression of PPAR-γ mRNA was determined by RT-PCR. RESULTS: Compared with the control group, Ang II (10-6 mol/L) caused a decrease in endothelial cell density, increased secretion of TNF-α, and decreased PPAR-γ mRNA expression. The serum containing Tianma Gouteng Decoction could inhibit the cell damage caused by AngII, reduce the secretion of TNF-α, and increase the expression of PPAR-γ mRNA. Conclusion: Tianma Gouteng Decoction can protect HUVECs from injury caused by AngII and protect endothelial cell function.
