目的:观察左、右归丸及其拆方对去卵巢大鼠肾脏TGFβ1/Smad1基因与蛋白表达的影响,并探讨补肾法对去卵巢模型大鼠肾脏TGFβ1/Smad1通路的影响。方法:将90只SD雌性大鼠随机分为空白组、假手术组、模型组、补佳乐组、左归丸组、右归丸组、共同药组、滋阴药组、补阳药组。除假手术、空白组外均进行双侧卵巢切除。经12周治疗分别以Q-PCR法、Westernblot法检测TGFβ1、TβRⅠ、TβRⅡ、Smad1在肾脏中基因和蛋白的表达。结果:与空白组相比,除假手术组外,TGFβ1、TβRⅠ、TβRⅡ、Smad1mRNA及蛋白表达降低(P<0.01);与模型组比较,各用药组TGFβ1、TβRⅠ、TβRⅡ、Smad1mRNA及蛋白表达升高(P<0.01);与补佳乐组比较,各用药组TGFβ1、TβRⅠ、TβRⅡ、Smad1表达低,其中左归丸组表达较高。结论:左、右归丸及其拆方可影响肾脏中TGFβ1/Smad1基因与蛋白的表达,其中以左归丸改变最为显著。
Objective: To observe the effects of Zuogui Gui Wan and its decomposed formulas on the expression of TGFβ1 / Smad1 gene and protein in the kidney of ovariectomized rats and to explore the effect of tonifying kidney on TGFβ1 / Smad1 pathway in the kidney of ovariectomized rats. Methods: Ninety SD female rats were randomly divided into blank group, sham operation group, model group, Bu Jiale group, Zuoguiwan group, Youguiwan group, common medicine group, Ziyin drug group, . In addition to sham surgery, both sides of the blank group were ovariectomized. The expression of TGFβ1, TβRI, TβRII and Smad1 in kidney were detected by Q-PCR and Western blot after 12 weeks of treatment. Results: Compared with the blank group, the expressions of TGFβ1, TβRⅠ, TβRⅡ and Smad1 mRNA and protein were significantly decreased (P <0.01). Compared with the model group, the expressions of TGFβ1, TβRⅠ, TβRⅡ, Smad1 mRNA and protein were increased (P <0.01). Compared with NGF group, the expression of TGFβ1, TβRⅠ, TβRⅡ and Smad1 in each drug group was low, among which Zuoguiwan group had higher expression. Conclusion: Zuogui Gui Wan and its disassembled formulas can affect the expression of TGFβ1 / Smad1 gene and protein in kidney, of which Zuo Gui Wan is the most obvious change.
