目的研究武汉市生活饮用水中有机提取物对人肝肿瘤HepG2细胞DNA损伤作用。方法用XAD-2/8复合树脂(XAD-2和XAD-8树脂等体积混合)等比富集武汉市生活饮用水中有机污染物,应用慧星试验检测有机提取物的遗传毒性。结果该市不同水源、不同水文期下饮用水中有机提取物对HepG2细胞均产生不同程度的DNA损伤作用。同一水文期不同水源比较,在剂量10和30ml/ml下,汉江水源末梢水有机提取物致DNA损伤程度大于长江水源(P<0.05);同一水源不同水文期时比较,在剂量10和30ml/ml下,枯水期有机提取物致DNA损伤程度大于丰水期(P<0.05)。结论武汉市主要水源汉江和长江的氯化饮用水有机提取物对HepG2细胞DNA均有损伤作用,损伤作用的严重程度从大到小为汉江水>长江水,枯水期>丰水期。
Objective To study the effect of organic extracts from drinking water in Wuhan city on DNA damage of human hepatoma HepG2 cells. Methods The organic contaminants in drinking water of Wuhan city were enriched in the same ratio of XAD-2/8 composite resin (XAD-2 and XAD-8 resin in equal volumes) and the genotoxicity of organic extracts was tested by Comet test. Results The HepG2 cells in different water sources and organic extracts of drinking water under different hydrological periods all had different degrees of DNA damage. Compared with different water sources in the same hydrological period, the DNA damage of the organic extracts from the water of the Hanjiang River water source was greater than that of the Yangtze River water source at the doses of 10 and 30ml / ml (P <0.05) ml, the degree of DNA damage induced by organic extract in dry season was higher than that in wet season (P <0.05). Conclusion The organic extracts of chlorinated drinking water, the main water sources in the Han River and the Yangtze River in Wuhan City, have the effect of damaging the DNA of HepG2 cells. The severity of the damage is from Hanjiang River to Yangtze River, dry season> wet season.
