目的:体外研究维甲类化合物的衍生物ATRA对A549肺癌细胞株作用,探讨ATRA对肿瘤防治作用的可能机制。方法:取对数生长期的常规培养的肺癌A549细胞用于试验,试验分1×10-5mol/L,1×10-6mol/L,1×10-7mol/L三组药物浓度和对照及空白对照5组,加药48h进行MTT测定,第6 d对1×10-5mol/L,1×10-6mol/L及对照组细胞进行流式细胞检测,并对1×10-5mol/L,1×10-6mol/L浓度进行电镜观察。结果:不同浓度ATRA对细胞增殖有明显地抑制作用,与阴性对照组比较,有统计学意义(P<0.01)。增殖抑制有良好剂量效应关系,(r=0.968),细胞阻滞于G0/G1期,高浓度组电镜下见凋亡细胞。结论:ATRA可诱导A549细胞分化及凋亡,为临床应用ATRA化学预防肺癌提供理论依据。
OBJECTIVE: To study the effect of retinoid ATRA derivatives on A549 lung cancer cell lines in vitro and to explore the possible mechanism of ATRA on tumor prevention and treatment. Methods: The lung cancer A549 cells cultured in logarithmic growth phase were used in the experiment. The test groups were divided into three groups: 1 × 10-5mol / L, 1 × 10-6mol / L, 1 × 10-7mol / L, The blank control group (5 groups) and the dosing group (48h) were used for MTT assay. Flow cytometry was performed on the 1 × 10-5mol / L, 1 × 10-6mol / L and control groups on the 6th day, and 1 × 10-5mol / L , 1 × 10-6mol / L concentration for electron microscopy. Results: Different concentrations of ATRA significantly inhibited cell proliferation, compared with the negative control group, with statistical significance (P <0.01). Proliferation inhibition has a good dose-response relationship, (r = 0.968), cell arrest in G0 / G1 phase, high concentration of apoptotic cells under electron microscopy. CONCLUSION: ATRA can induce A549 cell differentiation and apoptosis, providing a theoretical basis for clinical application of ATRA chemoprevention in lung cancer.
