目的建立云南省玉溪市甲型副伤寒沙门菌(SPA)分离株脉冲场凝胶电泳(PFGE)数据库,分析玉溪市各分子型别SPA的流行和分布情况,为本市SPA的监测和疫情的应急处理提供科学依据。方法采用SpeⅠ和XbaⅠ消化细菌染色体,对来自玉溪市6县1区194株SPA分离株进行PFGE分型和聚类分析。结果194株SPA中,XbaⅠ带型共有7种,分别为XbaⅠ01、XbaⅠ02、XbaⅠ06、XbaⅠ07、XbaⅠ09、XbaⅠ10、XbaⅠ19,其中XbaⅠ01为优势带型,占93.81%(182/194);SpeⅠ带型共有10种,分别为SpeⅠ01~10,其中SpeⅠ01、SpeⅠ02为优势带型,各占46.39%(90/194)和43.30%(84/194)。比较分析XbaⅠ带型和SpeⅠ带型,SpeⅠ酶切比XbaⅠ酶切具有更多的带型,主要是XbaⅠ01带型和XbaⅠ02带型可以分为更多的SpeⅠ带型,SpeⅠ01和SpeⅠ02带型均来自于XbaⅠ01带型。但是,也有不同的XbaⅠ带型归属于相同的SpeⅠ带型。结论建立了玉溪市SPAPFGE数据库,SpeⅠ酶切数据库与XbaⅠ酶切数据库具有较好的一致性。SpeⅠ01和SpeⅠ02或XbaⅠ01是玉溪流行的主要克隆,同时也不容忽视其他分子型别的危害。
Objective To establish the PFGE database of Salmonella paratyphi A (SPA) isolates from Yuxi City, Yunnan Province, and to analyze the prevalence and distribution of SPA of various molecular types in Yuxi city. Emergency treatment to provide a scientific basis. Methods The bacterial chromosomes were digested with Spe Ⅰ and Xba Ⅰ, and 194 SPA isolates from 6 districts in Yuxi city were genotyped by PFGE and clustered. Results Among 194 strains of SPA, there were 7 species of XbaⅠ, which were XbaⅠ01, XbaⅠ02, XbaⅠ06, XbaⅠ07, XbaⅠ09, XbaⅠ10 and XbaⅠ19, among which XbaⅠ01 was the dominant band, accounting for 93.81% (182/194) Species were SpeⅠ01 ~ 10, respectively. SpeⅠ01 and SpeⅠ02 were the dominant bands, accounting for 46.39% (90/194) and 43.30% (84/194), respectively. Compared with XbaⅠtype and SpeⅠtype, SpeⅠ digestion had more banding patterns than XbaⅠ digestion, mainly XbaⅠ01 banding and XbaⅠ02 banding could be divided into more SpeⅠ banding patterns. SpeⅠ01 and SpeⅠ02 banding patterns were all from In Xba Ⅰ 01 band type. However, there are also different XbaI patterns belonging to the same SpeI band pattern. Conclusion The database of SPAPFGE was established in Yuxi City. The Spe Ⅰ digestion database and the Xba Ⅰ digestion database had good consistency. SpeI01 and SpeI02 or XbaI01 are the main clones of Yuxi epidemic, but also can not ignore the harm of other molecular types.
