目的:研究黄芩提取物(SBE)对阿尔茨海默病(AD)模型小鼠的抗氧化作用。方法:ip给予D-半乳糖(150 mg/kg)和亚硝酸钠(100 mg/kg),每天1次,连续60 d以复制小鼠AD模型。60只小鼠随机均分为正常对照(等容氯化钠注射液)组、模型(等容氯化钠注射液)组、雌二醇(0.4 mg/kg)组与SBE高、中、低剂量(500、250、125 mg/kg)组,除正常对照组外,其余各组复制模型同时ig给药,每天1次,连续60 d。测定小鼠脑组织中超氧化物歧化酶(SOD)、过氧化氢酶(CAT)活性和丙二醛(MDA)含量,以及血清中谷胱甘肽过氧化物酶(GSH-Px)活性。结果:与正常对照组比较,模型组小鼠脑组织SOD、CAT活性减弱,MDA含量增加,血清中GSH-Px活性减弱,差异有统计学意义(P<0.01或P<0.05);与模型组比较,SBE高、中剂量组小鼠脑组织SOD、CAT活性增强,MDA含量减少,血清中GSH-Px活性增强,差异有统计学意义(P<0.01或P<0.05)。结论:SBE可增强AD模型小鼠抗氧化能力,其机制可能与改善脑组织与血清中相关抗氧化指标水平有关。
Objective: To study the anti-oxidative effect of skullcap extract (SBE) on Alzheimer’s disease (AD) model mice. METHODS: D-galactose (150 mg / kg) and sodium nitrite (100 mg / kg) were administered ip once daily for 60 days to replicate the mouse model of AD. 60 mice were randomly divided into normal control (sodium chloride injection) group, model group (isosmia sodium chloride injection group), estradiol (0.4 mg / kg) group and SBE high, middle and low Dose (500,250,125 mg / kg) group, except for the normal control group, the other groups were replicated ig administration mode, once a day for 60 days. The activity of superoxide dismutase (SOD), catalase (CAT) and the content of malondialdehyde (MDA) in the brain of mice were measured, and the activity of glutathione peroxidase (GSH-Px) in the serum was measured. Results: Compared with the normal control group, the activity of SOD and CAT in the model group decreased, the content of MDA increased and the activity of GSH-Px in the serum decreased, the difference was statistically significant (P <0.01 or P <0.05) Compared with SBE, the activity of SOD and CAT in brain tissue of high and middle dose SBE mice increased, the content of MDA decreased and the activity of GSH-Px in serum increased. The difference was statistically significant (P <0.01 or P <0.05). CONCLUSIONS: SBE can enhance the anti-oxidative ability of AD model mice, which may be related to the improvement of the level of anti-oxidative indexes in brain tissue and serum.
