从土党参中分离纯化多糖并进行体外抗氧化能力分析。试验通过微波辅助提取、乙醇沉淀、Sevag去蛋白质、透析及DEAE-Cellulose 52柱层析、Sephadex G-100柱层析分离纯化土党参多糖,采用比色法测定其含量。以Vc为对照,检测多糖组分对DPPH·、·OH和O_2~-·自由基的清除能力,得出土党参多糖单组分的体外抗氧化能力。试验结果:微波提取土党参多糖得率为12.58%,在4次脱蛋白后,蛋白脱出率达到55.2%,多糖的损失率为26.4%。经柱层析分离纯化得到2个含量较高的多糖组分CLP-1和CLP-2。它们均对DPPH·、·OH和O_2~-·自由基有一定抑制作用并表现出量效关系。
Isolation and Purification of Polysaccharides from Codonopsis Pilosula and Their Antioxidative Activities in. The microwave-assisted extraction, ethanol precipitation, Sevag deproteinization, dialysis, DEAE-Cellulose 52 column chromatography and Sephadex G-100 column chromatography were used to separate and purify the polysaccharides of Codonopsis pilosula. The contents were determined by colorimetry. Using Vc as control, the scavenging ability of polysaccharides to DPPH ·, · OH and O 2 - · · · ·· was detected, and the in vitro antioxidant activity of polysaccharides from Cortex Dactylon was obtained. The results showed that the yield of polysaccharide extracted from soil was 12.58%. After 4 times of deproteinization, the rate of proteolysis was 55.2% and the loss of polysaccharide was 26.4%. Two polysaccharides, CLP-1 and CLP-2, were isolated and purified by column chromatography. They both inhibited DPPH ·, · OH and O_2 ~ - · free radicals and showed a dose-response relationship.
