以新农1号狗牙根成熟种子为外植体,通过种子划破处理、不同激素浓度组合对种子发芽及愈伤组织诱导,不同继代培养基对愈伤组织增殖和质量影响的研究,结果表明:与未切割的新农1号狗牙根种子相比,切割后的新农1号狗牙根种子发芽率和愈伤组织诱导率均有显著提高,种子发芽率平均提高21%,愈伤组织诱导率平均提高14%,出愈时间提早5~10 d;愈伤组织诱导中,添加4.00 mg/L 2,4-D+1.00 mg/L 6-BA的诱导培养基所形成的愈伤组织质量较优;继代改造培养基中添加6%蔗糖+0.50 mg/L 2,4-D有利于胚性愈伤组织的生长,其分化率达50%。
The seedlings of mature bermudagrass of Xinnong 1 were used as explants. Seed germination and callus induction were induced by different combinations of seed concentration, and the effects of different subculture media on callus proliferation and quality were studied.
