目的 :通过研究 Ki- 6 7和 Bax基因产物在肾癌组织中的表达 ,探讨二者的相关性 ,以及 Ki- 6 7/Bax比率的意义。方法 :对 43例肾癌组织石蜡标本与 9例正常肾组织对照 ,采用 S- P法观察 Ki- 6 7与 Bax产物阳性表达及表达率的统计分析。结果 :Bax阳性表达定位于细胞浆 ,肾癌组织中阳性表达率 17.8% ,与正常肾组织 30 .2 %相比 ,差异十分显著 (P <0 .0 1)。但其表达与肿瘤分级、分期无关 (P >0 .0 5 ) ;Ki- 6 7阳性染色定位于细胞核 ,阳性表达与肿瘤恶性程度及预后密切相关 (P <0 .0 5 ) ,随肿瘤分级、分期的增高其阳性表达率升高。Ki- 6 7与 Bax的表达呈负相关 (r =- 0 .49,P <0 .0 5 )。在正常肾组织中 Ki- 6 7/Bax比率波动范围为 0 .5~ 2 .1,肾癌组织中为 0 .3~6 .2 ,二者比较差异显著 (P <0 .0 5 )。结论 :肾癌组织中 Ki- 6 7的阳性表达率可以作为肾癌恶性程度和预后的评价指标 ,Bax基因可能不参与肿瘤分化性行为 ,而是维持肿瘤生长的重要因素之一。 Ki- 6 7/Bax比率失调则可能是正常组织向肿瘤转化的原因之一。
OBJECTIVE: To investigate the relationship between the expression of Ki- 6 7 and Bax gene in renal cell carcinoma and the significance of Ki- 6 7 / Bax ratio. Methods: 43 cases of renal cell carcinoma specimens and 9 cases of normal kidney tissue contrast, S-P method was used to observe the expression of Ki- 6 7 and Bax products and statistical analysis of positive expression rate. Results: The positive expression rate of Bax in cytoplasm and renal cell carcinoma was 17.8%, which was significantly different from that in normal renal tissue (30.2%) (P <0.01). However, its expression was not associated with tumor grade and stage (P> 0.05). The positive expression of Ki- 6 7 localized in the nucleus, and the positive expression was closely related to the malignant degree and prognosis (P <0.05) , Staging increased its positive expression rate increased. Ki- 6 7 was negatively correlated with Bax expression (r = - 0.49, P <0.05). In normal kidney tissue, the ratio of Ki- 6 7 / Bax fluctuated in the range of 0.5-2.1 and in renal cell carcinoma ranged from 0.3-6.2, showing significant difference (P <0.05). Conclusion: The positive expression rate of Ki-67 in renal cell carcinoma can be used as the evaluation index of malignant degree and prognosis of renal cell carcinoma. Bax gene may not participate in tumor differentiation behavior, but it is one of the important factors to maintain tumor growth. Ki- 6 7 / Bax ratio imbalance may be one of the reasons for the normal tissue to tumor transformation.
