目的探讨脑肿瘤细胞增殖活性及凋亡与人类白细胞抗原-Dr(HLA-Dr)表达的相互关系。方法用免疫组化和原位凋亡细胞检测方法现察分析了44例原发性脑肿瘤。结果增殖细胞核抗原(PCNA)、Ki-67抗原(Ki-Dr)阳性细胞和凋亡细胞阳性率均为100%,29例(67.2%)表达HLA-Dr。PCNA和Ki-67阳性细胞密度及HLA-Dr阳性率和阳性细胞含量均随肿瘤恶性程度增高而升高,凋亡细胞密度随肿瘤恶性程度增高而减少。HLA-Dr表达水平与PCNA和Ki-67阳性细胞密度呈平行升高,HLA-Dr-、+、++、+++组间的凋亡细胞密度无显著性差异。结论本组结果提示以上四种指标均能较客观反映脑肿瘤的恶性程度,HLA-Dr表达水平与脑肿瘤细胞增殖速度密切相关,细胞增殖与凋亡失衡可能是脑肿瘤发生发展的重要环节。
Objective To investigate the relationship between the proliferation of human tumor cells and the expression of HLA-DR. Methods 44 cases of primary brain tumors were analyzed by immunohistochemistry and in situ detection of apoptotic cells. Results The positive rates of proliferating cell nuclear antigen (PCNA), Ki-67 positive cells and apoptotic cells were all 100%, and 29 cases (67.2%) expressed HLA-DR. The positive cell density of PCNA and Ki-67, the positive rate of HLA-DR and the content of positive cells increased with the increase of the malignant degree of the tumor. The density of apoptotic cells decreased with the increase of malignant degree of the tumor. The expression level of HLA-Dr was parallel to that of PCNA and Ki-67 positive cells. The density of apoptotic cells in HLA-Dr -, +, ++, +++ groups showed no significant difference. Conclusion The above results suggest that the above four indexes can objectively reflect the malignant degree of brain tumors. The expression level of HLA-Dr is closely related to the proliferation rate of brain tumor cells. The imbalance between cell proliferation and apoptosis may be an important link in the development and progression of brain tumors.
