目的:采用星点设计-效应面法优化千斤拔的回流提取工艺。方法:以乙醇浓度、提取时间和溶媒比为自变量,以染料木苷、染料木素和浸膏等的收率为因变量,通过对自变量的总评归一值进行多元线性回归和二项式拟合,采用效应面优化最佳的工艺条件。结果:各指标的二项式拟合方程均优于多元线性回归方程,确定回流提取工艺条件为乙醇浓度(X1)为60%~80%,溶媒比(X2)为24~26倍,提取时间(X3)为180~240 min,提取次数是2次。工艺验证的总评归一预测值与实际值的偏差为-7.2%,二项式拟合的复相关系数R=0.9343。结论:采用星点设计-效应面法优选方法优化的染料木苷和染料木素提取工艺,方法简便、预测性良好。
OBJECTIVE: To optimize the reflux extraction process by using the star point design-response surface method. Methods: The ethanol concentration, extraction time and solvent ratio were used as independent variables. The yield of genistin, genistein and extract was taken as the dependent variable. The multiple linear regression and binomial Fitting, using the effect surface to optimize the best process conditions. Results: The binomial fitting equation of each index was better than multivariate linear regression equation. The conditions of reflux extraction were determined as ethanol concentration (X1) 60% ~ 80%, solvent ratio (X2) 24 ~ 26 times, extraction time (X3) is 180 ~ 240 min, the number of extraction is 2 times. The deviation of the predicted total value of the process validation from the actual value is -7.2%, and the complex correlation coefficient of the binomial fit is R = 0.9343. Conclusion: The optimized genistein and genistein extraction process using the method of star-point design-response surface optimization is simple and predictive.
