目的研究三氯化铝(AlCl3)对雄性小鼠生殖细胞的遗传毒性。方法将20只清洁级健康昆明种雄性小鼠按体重随机分为4组,分别为低、中、高剂量AlCl3染毒组(染毒剂量分别为50、75、100mg/kg)和阴性对照组(0.9%生理盐水),每组5只。采用腹腔注射方式进行染毒,连续染毒2d,间隔1天,持续2周。观察染毒前后体重变化,并计算睾丸系数。采用彗星试验测定Olive尾矩,采用精子核荧光染色试验测定未成熟精子率。结果染毒前后高、中、低剂量AlCl3染毒组体重与阴性对照组比较,差异均无统计学意义(P>0.05)。高、中、低剂量AlCl3染毒组睾丸系数均小于阴性对照组,差异有统计学意义(P<0.05);且体重增长值和睾丸系数随着AlCl3染毒剂量的升高而减少。彗星试验中,高、中、低剂量AlCl3染毒组Olive尾矩均长于阴性对照组,差异有统计学意义(P<0.01);且Olive尾矩随着AlCl3染毒剂量的升高而增加。精子核荧光染色试验中,高、中、低剂量AlCl3染毒组未成熟精子率均高于阴性对照组,差异有统计学意义(P<0.01);且未成熟精子率随着AlCl3染毒剂量的升高而增加。结论AlCl3暴露可对雄性小鼠生殖细胞产生损伤。
Objective To study the genotoxicity of aluminum trichloride (AlCl3) on male mouse germ cells. Methods Twenty healthy Kunming male mice of clean level were randomly divided into 4 groups according to body weight: low, medium and high doses of AlCl3 (doses of 50, 75 and 100 mg / kg respectively) and negative controls (0.9% saline), 5 per group. Using intraperitoneal injection of exposure, continuous exposure 2d, 1 day interval, for 2 weeks. Body weight changes before and after exposure were observed, and testicular coefficient was calculated. Olive tail moment was measured by comet assay and immature sperm rate was determined by sperm nuclear fluorescent staining. Results There was no significant difference in body weight between high, medium and low doses of AlCl3 before and after exposure to the negative control group (P> 0.05). The testis coefficient of high, medium and low doses of AlCl3 was lower than that of the negative control group, the difference was statistically significant (P <0.05), and weight gain and testicular coefficient decreased with the increase of AlCl3 dose. In comet assay, the Olive tail moment of AlCl3-exposed group was longer than that of the negative control group (P <0.01), and Olive tail moment increased with the increase of AlCl3 dose. In the test of sperm nucleus fluorescence staining, the immature sperm rate in high, medium and low doses of AlCl3 was higher than that in negative control group (P <0.01), and the percentage of immature sperm with AlCl3 dose Increase and increase. Conclusion AlCl3 exposure can damage male germ cells.
