目的观察非诺贝特对肥胖大鼠脂联素蛋白多聚体构成的影响。方法 SD大鼠随机分为普通对照组(NC组)、高脂对照组(HF组)和非诺贝特组(FF组)。喂饲高脂饮食制作肥胖大鼠模型,以非诺贝特30mg·kg~(-1)·d~(-1)灌胃4周,ELISA法测定血清总脂联素,非还原非加热变性Western blot检测血清、皮下及内脏脂肪组织中各聚合态脂联素蛋白表达水平,计算高聚体(HMW)脂联素比例。结果 HF组大鼠血总脂联素、血清和内脏脂肪中HMW脂联素及HMW比例、皮下脂肪中HMW脂联素低于NC组。FF组血清、内脏脂肪中HMW脂联素及内脏脂肪中HMW比例高于HF组(P<0.05)。结论非诺贝特能提高内脏脂肪和血清中HMW脂联素蛋白水平。
Objective To observe the effect of fenofibrate on adiponectin multimer composition in obese rats. Methods SD rats were randomly divided into normal control group (NC group), high fat control group (HF group) and fenofibrate group (FF group). The model of obese rats was fed with high-fat diet, fed with fenofibrate 30 mg · kg -1 · d -1 for 4 weeks, and the serum total adiponectin, non-reducing non-heating degeneration Western blot was used to detect the level of adiponectin protein in serum, subcutaneous and visceral adipose tissue, and the ratio of HMW adiponectin was calculated. Results The total adiponectin, serum HMW adiponectin and HMW ratio in HF group and HMW adiponectin in subcutaneous fat group were lower than those in NC group. The HMW adiponectin and visceral fat in FF group were higher than HF group in serum and visceral fat (P <0.05). Conclusion Fenofibrate can increase HMW adiponectin protein in visceral fat and serum.
