目的:建立筛选抗真菌活性化合物的96孔板方法,筛选海洋活性菌株。方法:利用96孔板培养真菌孢子,将样品加入到真菌培养基中,在适宜条件下培养,通过显微镜来检测菌丝的生长情况,判断样品是否有抗真菌活性,筛选活性菌株。结果:通过对我国东海海洋微生物的筛选,从541株微生物中找到了25株具有抗黄瓜黑星病菌的活性菌株。结论:本生物检测模型方便、快速、费用低,并且可以进行高通量筛选、相关性好,是一种抗真菌天然产物的理想筛选模型。
Objective: To establish a 96-well plate method for screening antifungal active compounds to screen marine active strains. Methods: The fungal spores were cultured in 96-well plates. The samples were added to the fungal medium and cultured under appropriate conditions. The growth of mycelium was detected by microscopy to determine whether the samples had antifungal activity and the active strains were screened. Results: Twenty-five active strains of Culex pipiens from Culex pipiens pallens were identified from 541 microorganisms by screening marine microorganisms in the East China Sea. Conclusion: The bioassay model is convenient, rapid, low cost and can be used for high-throughput screening with good correlation. It is an ideal screening model of antifungal natural products.
