目的探讨石菖蒲有效成分β-细辛醚对皮质酮(CORT)诱导星形胶质细胞(AS)凋亡的干预作用。方法原代培养AS,经不同浓度β-细辛醚预处理后,加入CORT诱导AS损伤模型,采用MTT法检测AS的细胞活力,利用流式细胞术Annexin-V/PI法检测AS凋亡率,用实时定量RT-PCR技术检测Bax和Bcl-2表达水平。结果 CORT处理后,CORT组较空白对照组细胞活力低,凋亡率显著增加,Bax表达上调,而Bcl-2表达下调(P<0.05)。与CORT组比较,β-细辛醚组细胞活力高,凋亡率低,Bax的表达下调,Bcl-2表达上调(P<0.05)。结论β-细辛醚对CORT诱导的CORT凋亡具有抑制作用,下调Bax表达和上调Bcl-2表达可能是其作用的机制之一。
Objective To investigate the effect of β-asarone, an active ingredient of Acorus calamus, on the apoptosis of astrocyte (AS) induced by cortisone (CORT). Methods Primary cultured AS was pretreated with different concentrations of β-asarone, and then induced with CORT to induce AS injury. The cell viability was measured by MTT assay. The apoptosis rate of AS was detected by flow cytometry Annexin-V / PI assay , Real-time quantitative RT-PCR detection of Bax and Bcl-2 expression levels. Results After CORT treatment, the cell viability of CORT group was significantly lower than that of blank control group, the apoptosis rate was significantly increased, Bax expression was up-regulated and Bcl-2 expression was down-regulated (P <0.05). Compared with CORT group, β-asarone group had higher cell viability, lower apoptosis rate, lower Bax expression and higher Bcl-2 expression (P <0.05). Conclusion β-asarone can inhibit CORT-induced apoptosis of CORT cells. Down-regulation of Bax expression and up-regulation of Bcl-2 expression may be one of its mechanisms.
