目的:研究合欢皮总皂苷对人微血管内皮细胞(HMEC-1)增殖的影响。方法:采用人脐静脉微血管内皮细胞作为研究模型,经不同剂量合欢皮总皂苷作用24、48 h后,应用SRB法、HE染色法观察合欢皮总皂苷对HMEC-1细胞增殖、形态的影响;应用Hoechst染色法、扫描电镜、流式细胞仪PI单染方法分析合欢皮总皂苷对HMEC-1细胞生长的影响。结果:合欢皮总皂苷提取物具有抑制血管内皮细胞增殖的作用,其与时间、剂量均存在依赖关系,作用48 h的IC50为1.5μg/mL;合欢皮总皂苷使HMEC-1细胞周期阻滞在亚G0/G1期,高剂量作用48 h后可能引起细胞产生坏死。结论:合欢皮总皂苷通过影响细胞周期,部分引起细胞坏死,从而抑制人微血管内皮细胞增殖及血管形成。
Objective: To study the effect of total saponins of Acorus gramineus on the proliferation of human microvascular endothelial cells (HMEC-1). Methods: Human umbilical vein endothelial cells were used as a model to study the effects of total saponins of Acorus gramineus on the proliferation and morphology of HMEC-1 cells by different doses of total saponins of Acorus gramineus for 24,48 h. Hoechst staining, scanning electron microscopy and flow cytometry PI staining were used to analyze the effects of total saponins from Acorus gramineus on the growth of HMEC-1 cells. Results: The total aconitine extract could inhibit the proliferation of vascular endothelial cells, which was dependent on the time and dosage. IC50 was 1.5 μg / mL for 48 h. Total HGH inhibited the cell cycle arrest of HMEC-1 In sub-G0 / G1 phase, high-dose 48 h may cause cell necrosis. CONCLUSION: Acacia total saponin can inhibit the proliferation and angiogenesis of human microvascular endothelial cells by affecting the cell cycle and partly causing cell necrosis.
