自噬是维持正常细胞稳态所必需的一种动态自我降解和再循环过程,其最终目的是通过溶酶体蛋白酶降解底物,清除细胞内异常蛋白质和受损细胞器,维持细胞正常功能。越来越多的研究表明,病理情况下,神经细胞自噬紊乱与帕金森症的发生发展具有密切联系。论文综述了近年来自噬功能紊乱在帕金森症中的研究进展,介绍了帕金森症相关基因与自噬的关系,并讨论了靶向自噬的帕金森症治疗手段。
Autophagy is a dynamic process of self-degradation and recycling necessary to maintain normal cell homeostasis. The ultimate goal of autophagy is to degrade the substrate through lysosomal proteases, remove abnormal proteins and damaged organelles, and maintain the normal function of cells. More and more studies have shown that pathological conditions, neuronal autophagy disorder is closely linked with the occurrence and development of Parkinson’s disease. In this paper, the progress of autophagy dysfunction in Parkinson’s disease in recent years is reviewed. The relationship between Parkinson’s disease-related genes and autophagy is also introduced. The treatment of Parkinson’s disease that targets autophagy is also discussed.
