通过建立人工体外游离器官灌注系统对实验性异种移植免疫机制进行探讨,并结合免疫荧光病理及ELISA天然抗体测定法与该系统所观察结果作一综合分析,试图客观地评价此系统对异种移植实验研究的价值。本实验共分为五个组,A组为对照组(n=4),即用猪全血灌注猪肾脏;异种基因牛血灌注组为B组(n=4);C组为羊血灌注组(n=2);D组和E组分别为猴血(n=4)和人血组(n=4)。A组中,灌注持续200min均保持较低的肾血管阻力;B组在155min内仍保持较低的血管阻力;D组排斥发生非常迅速,仅4min;E组在55min时血管阻力明显增加;而C组在第70min时出现排斥反应。应用免疫荧光技术,D、E组均可见到IgM抗体沉积在肾小球间质内,B组见到较弱的IgM沉积,C组未得到好的结果。本实验中未见到IgG在超急排斥中起任何作用。ELISA法测定对照组IgM抗体水平变化不大。D组IgM吸收率最高(30.3%),E组为22.6%,B组吸收率最低(11.2%),C组为14.8%。灌注前后抗体IgM吸收率变化顺序为D、E、C、B组,与各实验组超急排斥反应的发生时间顺序一致。结论:本人工体外游离器官灌注系统是一种可靠的研究异种移植免?
