目的分析系统性红斑狼疮(Systemic lupus erythematosus,SLE)患者早期生长效应因子1(Early growth response1,EGR1)基因的表达,探讨SLE发病的分子机制。方法挑选位于SLE染色体连锁位点上的EGR1基因,利用实时荧光定量PCR技术,检测SLE活动期及相对静止期患者与健康对照组外周血单核细胞与T、B淋巴细胞亚群中EGR1的表达。结果 SLE患者外周血单核细胞和T淋巴细胞亚群中,EGR1基因的ΔCt值显著高于健康对照组(P<0.01),其表达明显下调;在B淋巴细胞亚群中,EGR1基因的ΔCt值略微升高,表达也呈下降趋势。在SLE患者外周血T淋巴细胞亚群中,EGR1基因的表达在活动期与相对静止期差异有统计学意义(P<0.05),活动期患者基因表达量下调趋势更为明显,SLEDAI积分与基因表达水平之间具有一定的相关性。结论 EGR1基因可能是位于5q31.1区段的一个SLE致病候选基因,且与疾病的活动性具有一定的相关性。
Objective To analyze the expression of early growth response 1 (EGR1) gene in patients with systemic lupus erythematosus (SLE) and to explore the molecular mechanism of SLE. Methods The EGR1 gene located at the SLE chromosomal linkage site was selected and the expression of EGR1 in peripheral blood mononuclear cells and T lymphocyte subsets of SLE patients and healthy patients was detected by real-time fluorescence quantitative PCR . Results The ΔCt of EGR1 gene in peripheral blood mononuclear cells and T lymphocyte subsets of SLE patients was significantly higher than that of healthy controls (P <0.01), and the expression of EGR1 gene was significantly down-regulated in SLE patients. In B lymphocyte subsets, Slightly higher value, the expression also showed a downward trend. The expression of EGR1 gene in peripheral blood T lymphocyte subsets of patients with SLE was significantly different between the active phase and the relative quiescent phase (P <0.05), while the expression of EGRE gene in the patients with active SLE was more significantly decreased There is a certain correlation between the expression levels. Conclusion The EGR1 gene may be a SLE pathogenic candidate gene located in the 5q31.1 region and has some correlation with disease activity.
