目的:观察益气化瘀方对大鼠前列腺增生组织血管新生和基质金属蛋白酶2(MMP-2)及其抑制物(TIMP-2)表达的影响。方法:50只SD大鼠去势7天后,皮下注射丙酸睾酮5mg.kg-1.d-1,同时给予益气化瘀方、非那雄胺混悬液灌胃,30天后股动脉放血处死大鼠,摘取前列腺,测量前列腺体积,计算前列腺指数(PI),检测前列腺微血管密度(MVD)和前列腺组织中MMP-2及其抑制物(TIMP-2)表达。结果:与正常组比较,模型组大鼠前列腺指数明显增大(P<0.01),前列腺组织MVD明显增高(P<0.01),MMP-2蛋白表达显著增高(P<0.01),MMP-2/TIMP-2比值明显增大(P<0.05);各给药组前列腺前列腺指数、MVD、MMP-2表达、MMP-2/TIMP-2比值均明显低于模型组(P<0.05或P<0.01);益气化瘀方中、高剂量组大鼠前列腺指数、MMP-2表达量、MMP-2/TIMP-2比值与非那雄胺组比较,差异均无统计学意义(P>0.05),但前列腺组织MVD明显低于非那雄胺组(P<0.05)。结论:中药益气化瘀方具有抗前列腺组织增生作用,其机制可能与抑制MMP-2过度表达,进而抑制前列腺增生组织新生血管形成有关。
Objective: To observe the effects of Yiqi Huayu Recipe on angiogenesis and expression of matrix metalloproteinase 2 (MMP-2) and its inhibitor (TIMP-2) in benign prostatic hyperplasia rats. Methods: Fifty SD rats were castrated for 7 days and injected with testosterone propionate 5 mg.kg-1.d-1 subcutaneously. The rats were given Yiqi Huayu Recipe and finasteride suspension for gavage. The femoral artery was excreted after 30 days The rats were sacrificed, the prostate was removed, the prostate volume was measured, the Prostate Index (PI) was calculated, the prostatic microvessel density (MVD) and the expression of MMP-2 and its inhibitor (TIMP-2) in prostate tissue were measured. Results: Compared with the normal group, the expression of MMP-2 and MMP-2 in the model group were significantly increased (P <0.01) (P <0.05 or P <0.05). Prostate index, MVD, MMP-2, MMP-2 / TIMP-2 ratio in each administration group were significantly lower than those in model group (P> 0.05). There was no significant difference in prostate index, MMP-2, MMP-2 / TIMP-2 ratio between the middle-dose and high-dose Yiqi Huayu Recipe groups and finasteride group , But the MVD of prostatic tissue was significantly lower than that of finasteride (P <0.05). Conclusion: Yiqi Huayu Recipe has anti-prostatic hyperplasia effect, and its mechanism may be related to the inhibition of over-expression of MMP-2 and the inhibition of neovascularization in benign prostatic hyperplasia.
