目的:研究露蜂房中蛋白成分抗白血病作用的机制,为露蜂房治疗白血病的临床应用和开发新的抗白血病药物提供实验和理论依据。方法:采用透射电子显微镜技术,观察露蜂房蛋白成分对白血病细胞(HL-60)的生长抑制作用和形态结构的影响,运用免疫组织化学方法和图像分析系统,研究露蜂房蛋白成分对HL-60细胞凋亡相关信号转导蛋白NF-κBp65、β-catenin及iNOS表达的影响。结果:透射电镜下发现露蜂房蛋白组白血病细胞呈典型的凋亡形态学改变。免疫组化显示,与生理盐水对照组比较,露蜂房蛋白组白血病细胞中NF-κBp65、β-catenin及iNOS表达减弱,NF-κBp65和β-catenin细胞核内转导减少。经图像分析检测和统计学处理后,均有显著性差异(P<0.05)。结论:露蜂房蛋白成分有明显抑制HL-60细胞的作用,其作用机制可能是通过调节凋亡相关信号传导因子NFκ-Bp65、β-catenin及iNOS的表达,从而诱导白血病细胞凋亡。
OBJECTIVE: To study the mechanism of anti-leukemia effect of protein components in dew-hives and to provide experimental and theoretical evidences for the clinical application of dew-hives in the treatment of leukemia and the development of new anti-leukemia drugs. Methods: Transmission electron microscopy (TEM) was used to observe the effect of protein composition of dewfly on the growth inhibition and morphological structure of leukemia cells (HL-60). Immunohistochemistry and image analysis system were used to study the effects of protein composition of HLB Effects of apoptosis related signal transduction proteins NF-κBp65, β-catenin and iNOS expression. Results: Transmission electron microscopy revealed a typical morphological changes of apoptotic cells in dew hives proteome leukemia cells. Immunohistochemistry showed NF-κBp65, β-catenin and iNOS expression decreased in NF-κBp65 and beta-catenin cells compared with saline control group. After image analysis and statistical analysis, there was a significant difference (P <0.05). CONCLUSION: The protein content of dewfly hives significantly inhibits the effect of HL-60 cells. Its mechanism may be through the regulation of the expression of apoptosis-related signal transduction factors NFκ-Bp65, β-catenin and iNOS to induce leukemia cell apoptosis.
