目的探讨外源性5’-核苷酸对大鼠酒精性肝损伤的影响。方法 60只雄性Wistar大鼠随机分成5组:空白对照组,酒精模型组,无核苷酸干预组,低、高剂量干预组。除空白对照组灌胃蒸馏水外,各组用50%酒精灌胃4w。处死空白对照组和酒精模型组大鼠,测定血清谷草转氨酶(AST)、谷丙转氨酶(ALT)、总甘油三酯(TG)、总胆固醇(TC)、高密度脂蛋白胆固醇(HDL-C)、白蛋白(ALB)、总蛋白(TP)、超氧化物歧化酶(SOD)和丙二醛(MDA)水平,观察肝脏病理改变;剩余3组大鼠分别给予无核苷酸饲料和添加低、高剂量5’-核苷酸饲料干预喂养2w,观察干预效果。结果酒精灌胃4w后大鼠肝体比(A/G)升高,血清ALT、AST、TC、HDL-C水平升高,肝组织出现轻微的病理改变,造模成功;无核苷酸饲料干预后,各异常指标都趋于正常,大鼠表现出良好的自我恢复能力;而5’-核苷酸饲料干预后,大鼠血清TG水平降低,ALB、A/G水平升高,SOD活力增加。结论添加外源性5’-核苷酸能够促进大鼠白蛋白合成,调节脂质代谢,增强其抗氧化能力,有助于肝组织损伤后修复。
Objective To investigate the effects of exogenous 5’-nucleotides on alcoholic liver injury in rats. Methods Sixty male Wistar rats were randomly divided into five groups: blank control group, alcohol model group, no nucleotide intervention group, low and high dose intervention group. In addition to blank control group intragastric administration of distilled water, each group with 50% alcohol gavage 4w. The rats in the blank control group and alcohol model group were sacrificed and the levels of serum AST, ALT, TG, TC, HDL-C were measured. (ALB), total protein (TP), superoxide dismutase (SOD) and malondialdehyde (MDA) were measured to observe the pathological changes of the liver. The remaining three groups were given no nucleotide diet and supplemented with low , High-dose 5’-nucleotide feed intervention feeding 2w, observe the effect of intervention. Results After 4 weeks of alcohol administration, the hepatic body ratio (A / G) increased, the levels of serum ALT, AST, TC and HDL-C increased and the pathological changes of liver tissue were slight. After the intervention, all the abnormal indexes tended to be normal, and the rats showed good self-recovery ability. After the intervention of 5’-nucleotide diet, the levels of serum TG, ALB and A / G were increased, while SOD activity increase. Conclusion The addition of exogenous 5’-nucleotides can promote rat albumin synthesis, regulate lipid metabolism, enhance its anti-oxidative capacity, and help to repair liver injury.
